    Selective targeting of proteins by hybrid polyoxometalates: Interaction between a bis-biotinylated hybrid conjugate and avidin

    The Keggin-type polyoxometalate [\u3b3-SiW10O36]8 12 was covalently modified to obtain a bis-biotinylated conjugate able to bind avidin. Spectroscopic studies such as UV-vis, fluorimetry, circular dichroism, coupled to surface plasmon resonance technique were used to highlight the unique interplay of supramolecular interactions between the homotetrameric protein and the bis-functionalized polyanion. In particular, the dual recognition mechanism of the avidin encompasses (i) a complementary electrostatic association between the anionic surface of the polyoxotungstate and each positively charged avidin subunit and (ii) specific host-guest interactions between each biotinylated arm and a corresponding pocket on the tetramer subunits. The assembly exhibits peroxidase-like reactivity and it was used in aqueous solution for L-methionine methyl ester oxidation by H2O2. The recognition phenomenon was then exploited for the preparation of layer-by-layer films, whose structural evolution was monitored in situ by ATR-FTIR spectroscopy.     Factors influencing weight loss practices in Italian boxers: A cluster analysis

It is common practice in combat sports that athletes rapidly lose body weight before a match, by applying different practices—some safer and others possibly dangerous. The factors behind the choice of practices utilised have not been fully studied. This study aimed to investigate the weight loss strategies used by Italian boxers and to look at the difference between higher and lower risk practice adaptors. A modified version of a validated questionnaire has been sent to 164 amateur (88%) and professional (12%) boxers by email. A heatmap with hierarchical clustering was used to explore the presence of subgroups. Weight loss strategies were used by 88% of the athletes. Two clusters were found, defined by the severity of weight loss behaviours. Professional fighters, high-level athletes and females were more represented in Cluster 2, the one with more severe weight-loss practices. These athletes were characterised by a higher weight loss magnitude and frequency throughout the season and reported being more influenced by physicians and nutritionists, compared with the boxers in Cluster 1. Not all the weight loss practices are used with the same frequency by all boxers. The level of the athlete and the boxing style have an influence on the weight-cutting practices

    Berkeley 29, the most distant old open cluster

    We present CCD BVI photometry of the old open cluster Berkeley 29, located in the anticentre direction. Using the synthetic Colour - Magnitude Diagrams technique we estimate at the same time its age, reddening, distance, and approximate metallicity using three types of stellar evolutionary tracks. The best solutions give: age=3.4 or 3.7 Gyr, (m-M)_0 = 15.6 or 15.8 with E(B-V) = 0.13 or 0.10, and metallicity lower than solar (Z=0.006 or 0.004), depending on the adopted stellar models. Using these derived values, Be 29 turns out to be the most distant open cluster known, with Galactocentric distance R_GC= 21.4 to 22.6 kpc. Hence, Be 29 qualitatively follows both the age-metallicity relation and the metal abundance gradient typical of Galactic disc objects.     Metabolic and molecular rearrangements of Sauvignon Blanc (Vitis vinifera L.) berries in response to foliar applications of specific dry yeast

    Dry yeast extracts (DYE) are applied to vineyards to improve aromatic and secondary metabolic compound content and wine quality; however, systematic information on the underpinning molecular mechanisms is lacking. This work aimed to unravel, through a systematic approach, the metabolic and molecular responses of Sauvignon Blanc berries to DYE treatments. To accomplish this, DYE spraying was performed in a commercial vineyard for two consecutive years. Berries were sampled at several time points after the treatment, and grapes were analyzed for sugars, acidity, free and bound aroma precursors, amino acids, and targeted and untargeted RNA-Seq transcriptional profiles. The results obtained indicated that the DYE treatment did not interfere with the technological ripening parameters of sugars and acidity. Some aroma precursors, including cys-3MH and GSH3MH, responsible for the typical aromatic nuances of Sauvignon Blanc, were stimulated by the treatment during both vintages. The levels of amino acids and the global RNA-seq transcriptional profiles indicated that DYE spraying upregulated ROS homeostatic and thermotolerance genes, as well as ethylene and jasmonic acid biosynthetic genes, and activated abiotic and biotic stress responses. Overall, the data suggested that the DYE reduced berry oxidative stress through the regulation of specific subsets of metabolic and hormonal pathway

    Old open clusters and the Galactic metallicity gradient: Berkeley 20, Berkeley 66, and Tombaugh 2

    To study the crucial range of Galactocentric distances between 12 and 16 kpc, where little information is available, we have obtained VI CCD imaging of Berkeley 20 and BVI CCD imaging of Berkeley 66 and Tombaugh 2, three distant, old open clusters. Using the synthetic colour magnitude diagram (CMD) technique with three types of evolutionary tracks of different metallicities, we have determined age, distance, reddening and indicative metallicity of these systems. The CMD of Be 20 is best reproduced by stellar models with a metallicity about half of solar (Z=0.008 or 0.01), in perfect agreement with high resolution spectroscopic estimates. Its age is between 5 and 6 Gyr from stellar models with overshooting and between 4.3 and 4.5 Gyr from models without it. The distance modulus from the best fitting models is always (m-M)0=14.7 (corresponding to a Galactocentric radius of about 16 kpc), and the reddening E(B-V) ranges between 0.13 and 0.16. A slightly lower metallicity (Z ~ 0.006) appears to be more appropriate for Be 66. This cluster is younger, (age of 3 Gyr), and closer, (m-M)0=13.3 (i.e., at 12 kpc from the Galactic centre), than Be 20, and suffers from high extinction, 1.2 < E(B-V) < 1.3, variable at the 2-3 per cent level. Finally, the results for To 2 indicate that it is an intermediate age cluster, with an age of about 1.4 Gyr or 1.6-1.8 Gyr for models without and with overshooting, respectively. The metallicity is about half of solar (Z=0.006 to 0.01), in agreement with spectroscopic determinations. The distance modulus is (m-M)0=14.5, implying a distance of about 14 kpc from the Galactic centre; the reddening E(B-V) is 0.31-0.4, depending on the model and metallicity, with a preferred value around 0.34.Comment: 19 pages, 19 figures, 8 tables Accepted 2010 November 5.     Laboratory testing in the emergency department: an Italian Society of Clinical Biochemistry and Clinical Molecular Biology (SIBioC) and Academy of Emergency Medicine and Care (AcEMC) consensus report

    The mainstay of patient-oriented laboratory testing in emergency settings entails selecting number and type of tests according to valid criteria of appropriateness. Since the pattern of urgent tests requesting is variable across different institutions, we designed a joined survey between the Academy of Emergency Medicine and Care (AcEMC) and the Italian Society of Clinical Biochemistry and Clinical Molecular Biology (SIBioC) for reaching tentative consensus about the most informative diagnostic tests in emergency settings. A survey, containing the most commonly performed urgent laboratory tests and the relative clinical indications, was disseminated to eight relevant members of AcEMC and eight relevant members of SIBioC. All contributors were asked to provide numerical scores for the different laboratory parameters, where 1 indicated strongly recommended, 2 recommended in specific circumstances, and 3 strongly discouraged. The mean results of the survey were presented as the mean of responders' values, and the parameters were finally classified as strongly recommended (mean value, 1.0-1.5), somehow recommended (mean value, 1.5-2.0), discouraged (mean value, 2.0-2.5) and strongly discouraged (mean value, 2.5-3.0). The results of the survey allowed defining a hierarchy of priority, wherein 24 tests were strongly recommended. The use of 5 common tests was instead strongly discouraged. For 16 additional parameters in the list, the consensus ranged between somehow recommended and discouraged. We hope that results presented in this joint AcEMC-SIBioC consensus document may help harmonizing panel of tests and requesting patters in emergency setting, at least at a national level

    DETECTION OF WIDESPREAD HYDRATED MATERIALS ON VESTA BY THE VIR IMAGING SPECTROMETER ON BOARD THE DAWN MISSION

    Water plays a key role in the evolution of terrestrial planets, and notably in the occurrence of Earth's oceans. However, the mechanism by which water has been incorporated into these bodies—including Earth—is still extensively debated. Here we report the detection of widespread 2.8 μm OH absorption bands on the surface of the asteroid Vesta by the VIR imaging spectrometer on board Dawn. These observations are surprising as Vesta is fully differentiated with a basaltic surface. The 2.8 μm OH absorption is distributed across Vesta's surface and shows areas enriched and depleted in hydrated materials. The uneven distribution of hydrated mineral phases is unexpected and indicates ancient processes that differ from those believed to be responsible for OH on other airless bodies, like the Moon. The origin of Vestan OH provides new insight into the delivery of hydrous materials in the main belt and may offer new scenarios on the delivery of hydrous minerals in the inner solar system, suggesting processes that may have played a role in the formation of terrestrial planets

    COVID-19 in rheumatic diseases in Italy: first results from the Italian registry of the Italian Society for Rheumatology (CONTROL-19)

    OBJECTIVES: Italy was one of the first countries significantly affected by the coronavirus disease 2019 (COVID-19) epidemic. The Italian Society for Rheumatology promptly launched a retrospective and anonymised data collection to monitor COVID-19 in patients with rheumatic and musculoskeletal diseases (RMDs), the CONTROL-19 surveillance database, which is part of the COVID-19 Global Rheumatology Alliance. METHODS: CONTROL-19 includes patients with RMDs and proven severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) updated until May 3rd 2020. In this analysis, only molecular diagnoses were included. The data collection covered demographic data, medical history (general and RMD-related), treatments and COVID-19 related features, treatments, and outcome. In this paper, we report the first descriptive data from the CONTROL-19 registry. RESULTS: The population of the first 232 patients (36% males) consisted mainly of elderly patients (mean age 62.2 years), who used corticosteroids (51.7%), and suffered from multi-morbidity (median comorbidities 2). Rheumatoid arthritis was the most frequent disease (34.1%), followed by spondyloarthritis (26.3%), connective tissue disease (21.1%) and vasculitis (11.2%). Most cases had an active disease (69.4%). Clinical presentation of COVID-19 was typical, with systemic symptoms (fever and asthenia) and respiratory symptoms. The overall outcome was severe, with high frequencies of hospitalisation (69.8%), respiratory support oxygen (55.7%), non-invasive ventilation (20.9%) or mechanical ventilation (7.5%), and 19% of deaths. Male patients typically manifested a worse prognosis. Immunomodulatory treatments were not significantly associated with an increased risk of intensive care unit admission/mechanical ventilation/death. CONCLUSIONS: Although the report mainly includes the most severe cases, its temporal and spatial trend supports the validity of the national surveillance system. More complete data are being acquired in order to both test the hypothesis that RMD patients may have a different outcome from that of the general population and determine the safety of immunomodulatory treatments

    What is the role of the placebo effect for pain relief in neurorehabilitation? Clinical implications from the Italian consensus conference on pain in neurorehabilitation

    Background: It is increasingly acknowledged that the outcomes of medical treatments are influenced by the context of the clinical encounter through the mechanisms of the placebo effect. The phenomenon of placebo analgesia might be exploited to maximize the efficacy of neurorehabilitation treatments. Since its intensity varies across neurological disorders, the Italian Consensus Conference on Pain in Neurorehabilitation (ICCP) summarized the studies on this field to provide guidance on its use. Methods: A review of the existing reviews and meta-analyses was performed to assess the magnitude of the placebo effect in disorders that may undergo neurorehabilitation treatment. The search was performed on Pubmed using placebo, pain, and the names of neurological disorders as keywords. Methodological quality was assessed using a pre-existing checklist. Data about the magnitude of the placebo effect were extracted from the included reviews and were commented in a narrative form. Results: 11 articles were included in this review. Placebo treatments showed weak effects in central neuropathic pain (pain reduction from 0.44 to 0.66 on a 0-10 scale) and moderate effects in postherpetic neuralgia (1.16), in diabetic peripheral neuropathy (1.45), and in pain associated to HIV (1.82). Moderate effects were also found on pain due to fibromyalgia and migraine; only weak short-term effects were found in complex regional pain syndrome. Confounding variables might have influenced these results. Clinical implications: These estimates should be interpreted with caution, but underscore that the placebo effect can be exploited in neurorehabilitation programs. It is not necessary to conceal its use from the patient. Knowledge of placebo mechanisms can be used to shape the doctor-patient relationship, to reduce the use of analgesic drugs and to train the patient to become an active agent of the therapy
